四、Standard Usage Method


Step 1: Equipment Installation and Inspection

Place the microtome on a flat and stable experimental bench to ensure the equipment is placed horizontally without shaking. Check whether all components of the equipment (sample clamp, knife holder, handwheel, digital display system, etc.) are intact, the blade is installed firmly, the handwheel lock function is normal, and the equipment is well grounded.


Step 2: Sample and Blade Installation

Fix the paraffin-embedded tissue block on the sample clamp, tighten the wax block clamp and angle adjustment bolt, adjust the sample azimuth angle to ensure the sample is parallel to the blade. Select a suitable blade (disposable/reusable), install it on the knife holder, tighten the knife holder locking handle, adjust the knife holder position to ensure the blade is sharp and installed firmly.


Step 3: Parameter Setting

Set the section thickness (0-25μm) and trimming thickness (0-500μm) through the thickness dial, and confirm that the parameters displayed on the eLCD screen are correct. Adjust the knife holder position and sample feed according to the sample type to ensure the slicing parameters meet the experimental requirements.

Step 4: Slicing Operation

Unlock the handwheel, rotate the handwheel at a constant speed for trimming. After the sample surface is flat, switch to the set section thickness and start continuous sectioning. During the slicing process, check the slice count in real time through the digital display screen to ensure the slices are uniform and wrinkle-free; if abnormal slicing occurs, stop the operation immediately and check the blade and sample position.


Step 5: Operation Completion and Equipment Maintenance

After slicing is completed, lock the handwheel, remove the sample and blade, clean the blade and equipment surface, and remove residual paraffin and tissue debris. Reset the equipment parameters, turn off the power, perform equipment cleaning and maintenance, and regularly lubricate the transmission components to extend the service life of the equipment.
